They were both allegedly gunned down out the front of their home on Salisbury Street in Yarraville.
Emergency services were called to the home about 10.30am on Tuesday, where they found the couple.
Mrs Musai had died at the scene, and Mr Musai was rushed to hospital in a critical condition.
Police said the pair had both been shot in the upper body, and allege a firearm was found at the scene.
Another man, a 55-year-old from Altona North, was taken to hospital under police guard with what appeared to be gunshot wounds.
